Found this timely; I've just done similar but with a surface flat, offset depth gauge, and a distance piece. As shown in the OPs image, you're trying to measure the depth from the seating plane to the glass, and from the seating plane to the film. Given that there can be all sorts of lumpy bits on the back of the glass holder, it can be tricky to mount; I like the Jim's quick and easy gauge.
My MPP was designed to take glass plates, not the modern film holder, and while it's fine with a home-made film carrier plate in a glass plate holder, it isn't quite right for a film holder. Standards change, with time.
(Another way to measure it is to use a steel rule instead of the spirit level; rock it across its width to find the deep point).
