What matters is that the long side of the film is parallel with the front of the scanner. At least it does if you want the max optical resolution that the scanner is capable of.

I use the hold down plate the normal way, not because it only works that way. as you have found it works longways down the bed works too. I use it as an alignment device so I do not have to ever rotate in Photoshot, which is a real micro resolution smearer.

I also have a complete frame and plate system for all major formats.
I got lucky, since the seller was local and I spent lots of time with him, he was very generous with the accessories, including extra lamps etc.

I tried some experiments with wet mounting. The mask is just a black cardboard customised for the 5x4. I don t have one like yours in the photo!
A couple of thoughts
The mask can also go on top of your hold down plate. The ones above in the picture are a plastic vinyl. Screen sells them btw.
I would be careful with what material comes in contact with mounting fluid. The bed and plate are a proper acrylic, but I would be wary of other materials of unknown origin. It's solvent action may cause other materials to be deposited on your film. It may be overkill but you never know?
