In a properly designed combination of camera and film holder, the position of the ground glass when there is no holder in the camera is supposed to be exactly the same as the position of the film when the holder is in the camera.
Therefore, when you focus on the ground glass, you are causing the image to be in focus on what will eventually be the film plane when the holder is inserted.

As has already been pointed out, if the camera is properly set upt, the position of the gg and that of t he film should be identical to within certain tolerances. But this may not always be the case for a given camera. You can check this as follows. Tape something with lots of fine detail, such as the classfied section of a newspaper, to a wall. Set up the camera at about six to eight feet from the wall, pointed at a 45 degree angle, Make sure the camera is level, Make a mark in the center of the filed on the target and focus on it as best you can. Put in a loaded film holder and take a picture wide open. Repeat this several times. Examine the developed film. You should see a clear area around where you focused. If the camera is adjusted properly it shold be roughly centered on the focus mark. This may differ a bit from one shot to another, but if it is consistently off in all your shots, your camera needs adjusting.
